以2-氯-4-氰基吡啶為原料合成4-氰基-2-氟吡啶的一般步驟:將2-氯-4-氰基吡啶(6.0 g,43.5 mmol)、氟化鉀(7.56 g,130.3 mmol)和溴化四丁基鏻(14.8 g,43.7 mmol)溶于1-甲基-2-吡咯烷酮(20 mL)中,加熱至100°C反應(yīng)18小時。反應(yīng)完成后,用水稀釋反應(yīng)混合物,并用乙酸乙酯(EtOAc)萃取。有機相依次用水和鹽水洗滌,無水硫酸鈉(Na2SO4)干燥,減壓濃縮,得到目標產(chǎn)物4-氰基-2-氟吡啶(2.3 g,收率43%)。質(zhì)譜(ES)顯示m/z 123.1([M + H]+)。核磁共振氫譜(400 MHz,CDCl3)δ 8.43(d,1H,J = 5.2 Hz),7.45(m,1H),7.22(m,1H)。
