以3-氯-6-甲基噠嗪為原料合成3-肼基-6-甲基噠嗪的一般步驟:[參考例35] 將一水合肼(45 mL)加入至3-氯-6-甲基噠嗪(3.00 g)的乙醇(45 mL)懸浮液中,所得混合物加熱回流2.5小時(shí)。反應(yīng)完成后,將混合物在空氣中冷卻,隨后通過(guò)減壓蒸發(fā)去除溶劑,得到殘余物。該殘余物通過(guò)硅膠柱色譜法純化,使用氯仿-甲醇-水(7:3:1,v/v)的下層溶劑作為洗脫劑,最終獲得標(biāo)題化合物3-肼基-6-甲基噠嗪(2.35 g,產(chǎn)率81%),為固體。產(chǎn)物結(jié)構(gòu)經(jīng)1H-NMR(400 MHz, DMSO-d6)確認(rèn):δ 2.39(3H, s), 4.20(2H, br), 6.94(1H, d, J = 9.3 Hz), 7.18(1H, d, J = 9.3 Hz), 7.64(1H, br)。ESI-MS m/z:125([M + H]+)。
