以4-環(huán)己酮羧酸和叔丁醇為原料合成4-氧代環(huán)己烷羧酸叔丁酯的一般步驟:向中間體7(4-環(huán)己酮羧酸,5.0g,35mmol)在吡啶(19mL)和叔丁醇(27mL)中的冰冷溶液中逐滴加入三氯氧磷(POCl3,4.7mL,50.6mmol)。將反應(yīng)混合物緩慢升溫至室溫并持續(xù)攪拌4小時(shí)。反應(yīng)完成后,將粗反應(yīng)混合物小心倒入冰水中,并用乙酸乙酯(EtOAc)進(jìn)行萃取。合并有機(jī)層,用飽和食鹽水洗滌,無水硫酸鈉(Na2SO4)干燥,過濾后減壓濃縮,得到中間體8(4-氧代環(huán)己烷羧酸叔丁酯,4.0g,收率58%),該產(chǎn)物無需進(jìn)一步純化即可用于下一步反應(yīng)。產(chǎn)物經(jīng)1H NMR(400MHz,CDCl3)確認(rèn):δ 2.66(tt,J = 9.6, 3.9Hz,1H),2.48(dt,J = 14.8, 5.4Hz,2H),2.36(m,2H),2.18(ddd,J = 14.1, 8.7, 4.4Hz,2H),2.01(dtd,J = 14.4, 9.5, 4.8Hz,2H),1.48(s,9H)。LC/MS分析顯示:m/z計(jì)算值198.3,實(shí)測值199.1(M + 1)+。
