以2,4-二羥基-6-甲基吡啶-5-羧酸甲酯為原料合成2,4-二氯-6-甲基嘧啶-5-羧酸甲酯的一般步驟:將6-甲基-2,4-二氧代-1,2,3,4-四氫嘧啶-5-甲酸甲酯(1g,5.43mmol)懸浮于POCl3(10mL)中,加入N,N-二甲基苯胺(10滴)。將反應(yīng)混合物在105℃下加熱6小時,直至形成澄清溶液。反應(yīng)完成后,將混合物冷卻至室溫,減壓濃縮,隨后倒入冰水中,并用乙酸乙酯(EtOAc)萃取。合并有機(jī)相,依次用水和飽和鹽水洗滌,經(jīng)無水硫酸鈉干燥后,減壓濃縮,得到黃綠色固體2,4-二氯-6-甲基嘧啶-5-羧酸甲酯(940mg,收率78%)。質(zhì)譜分析:[M + H]+ C7H6Cl2N2O2計算值為222,實(shí)測值為221、223。
