(步驟1)向含有濃縮的3-氨基吡啶-2-甲酸(20g,144.9mmol)的甲醇(300mL)溶液中緩慢加入硫酸(5.4mL)。將反應(yīng)混合物加熱至回流狀態(tài),持續(xù)反應(yīng)5天。反應(yīng)完成后,通過減壓蒸餾濃縮反應(yīng)混合物。隨后,向濃縮物中加入水,并逐滴加入碳酸鈉直至不再觀察到氣泡產(chǎn)生。使用二氯甲烷(每次等量,共三次)對混合物進(jìn)行萃取。合并有機(jī)層,并用無水硫酸鈉干燥。最后,通過減壓蒸發(fā)去除溶劑,得到3-氨基吡啶-2-羧酸甲酯(17.0g,產(chǎn)率77%),產(chǎn)物為淺黃色固體。1H-NMR(300MHz,CDCl3)數(shù)據(jù)如下:δ3.95(3H,s),5.72(2H,brs),7.03(1H,dd,J = 0.99,8.35Hz),7.18-7.21(1H,m),8.05(1H,DD,J =0.96,4.04Hz)。
