在0℃條件下,向攪拌中的5-氨基煙酸(10.0g,72.5mmol)甲醇溶液(100mL)中緩慢滴加SOCl2(10.4g,86.9mmol)。將反應(yīng)混合物逐漸升溫至室溫后,繼續(xù)回流反應(yīng)16小時(shí)。反應(yīng)完成后,冷卻混合物,并在真空條件下濃縮。殘余物用去離子水(200mL)稀釋,隨后用NaHCO3水溶液中和至pH=7。水相混合物用二氯甲烷(DCM,100mL×2)進(jìn)行萃取。合并有機(jī)層,用飽和食鹽水(100mL×2)洗滌,無水Na2SO4干燥,過濾后,將濾液在真空條件下濃縮至干,得到白色固體5-氨基吡啶-3-羧酸甲酯9.5g,產(chǎn)率86%。產(chǎn)物經(jīng)1H NMR(DMSO-d6, 400MHz)表征:δ=8.24(1H, d),8.12(1H, d),7.42(1H, dd),5.65(2H, brs),3.84(3H, s)。
