以N-甲基哌嗪和對氟苯腈為原料合成4-(4-甲基哌嗪-1-基)苯甲腈的一般步驟如下:向含有4-氟芐腈(1.0g,8.25mmol)和碳酸鉀(2.27g,16.51mmol)的二甲基亞砜(7mL)溶液中加入1-甲基哌嗪(1.36mL,12.38mmol)。反應混合物在室溫下攪拌反應。反應完成后,通過常規(guī)后處理得到目標產(chǎn)物4-(4-甲基哌嗪-1-基)苯甲腈(1.54g,收率93%)。產(chǎn)物結(jié)構(gòu)通過1H-NMR(500MHz,CDCl3)和ESI-MS進行確認:1H-NMR δ 2.36(3H,s),2.55(4H,t,J = 4.88Hz),3.35(4H,t,J = 4.88Hz),6.87(2H,d,J = 8.78Hz),7.49(2H,d,J = 8.78Hz);ESI-MS m/z 202.1([M+H]+)。
