以甲醇和2,4-二氨基-6-氯嘧啶為原料合成2,4-二氨基-6-甲氧基嘧啶的一般步驟:向裝有320 mL甲醇的燒瓶中加入9.2 g(0.40 mol)金屬鈉,攪拌至完全溶解。隨后,向此溶液中加入28.9 g(0.20 mol)2,4-二氨基-6-氯嘧啶。將反應(yīng)混合物置于回流條件下,持續(xù)攪拌44小時。反應(yīng)完成后,通過過濾移除沉淀的氯化鈉。將濾液濃縮至干燥,得到粗產(chǎn)物。將粗產(chǎn)物溶于300 mL乙醇中,加熱至沸騰后進(jìn)行熱過濾。濾液靜置過夜,使2,4-二氨基-6-甲氧基嘧啶結(jié)晶析出。通過母液的重結(jié)晶可進(jìn)一步純化產(chǎn)物。干燥后,得到白色固體狀的2,4-二氨基-6-甲氧基嘧啶(58.8 g,收率84%)。熔點:166-169℃。1H-NMR(300 MHz, d6-DMSO):δ= 3.18(s, 3H, 6-OMe),5.06(s, 1H, 5-H),5.94(s, 2H, NH2),6.05(s, 2H, NH2)。13C-NMR(125 MHz, d6-DMSO):δ= 52.8(6-OMe),76.1(5-C),163.3(2-C),166.3(4-C),170.8(6-C)。
