以3-氯-5-硝基甲苯為原料合成3-氯-5-甲基苯胺的一般步驟:向含有1-氯-3-甲基-5-硝基苯(5.0 g,29 mmol)的乙醇溶液(75 mL)中加入二水合氯化亞錫(SnCl2·2H2O,32.8 g,146 mmol)。將反應(yīng)混合物加熱回流3.0小時(shí)。反應(yīng)完成后,將溶液在減壓下濃縮,殘余物重新溶解于氫氧化鈉水溶液中,過濾,并用乙酸乙酯(EtOAc)萃取。合并有機(jī)層,用飽和食鹽水洗滌,無(wú)水硫酸鎂(MgSO4)干燥,減壓濃縮。粗產(chǎn)物通過硅膠柱色譜法純化,得到3-氯-5-甲基苯胺(4.0 g,收率97%)為淺黃色固體。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(500 MHz,CDCl3)和ESI-MS確認(rèn):1H NMR δ 6.56(s,1H),6.48(s,1H),6.36(s,1H),3.66(bs,2H),2.23(s,3H);ESI-MS:m/z 141.7([M + H]+)。
