一般步驟:在室溫下,向攪拌的1,4-二氮雜雙環(huán)[3.2.2]壬-3-酮(1.0 g,7.2 mmol)的1,4-二惡烷(7.2 mL)溶液中緩慢加入氫化鋁鋰[2.0 M in THF](4.1 mL,8.2 mmol)。隨后,將反應(yīng)混合物加熱至回流狀態(tài),維持6小時(shí),然后冷卻至室溫。通過逐步添加水(200 μL)、15% NaOH水溶液(200 μL)和水(600 μL)來淬滅反應(yīng)。將混合物通過硅藻土墊過濾,并用乙酸乙酯(EtOAc)洗滌。合并濾液后,在真空下濃縮,得到1,4-二氮雜雙環(huán)[3.2.2]壬烷(0.82 g,收率90%),該產(chǎn)物無需進(jìn)一步純化即可用于后續(xù)反應(yīng)。產(chǎn)物的1H NMR(400 MHz,CDCl3)數(shù)據(jù)如下:δ 3.28-3.25(m,1H),2.99-2.95(m,8H),1.86-1.80(m,3H),1.69-1.64(m,2H)ppm。
