一般步驟:向N-羥基琥珀酰亞胺(640.3 mg,5.56 mmol)的氯仿(8.5 mL)溶液中加入三乙胺(861.6 μL,6.18 mmol),于0℃下攪拌。隨后,在5分鐘內(nèi)緩慢滴加α-氯乙酰氯,并在0℃下繼續(xù)攪拌20分鐘。反應(yīng)完成后,用冰冷的水(15 mL)和鹽水(15 mL)洗滌反應(yīng)混合物,隨后在減壓條件下濃縮至體積為1.7 mL。濃縮液經(jīng)無(wú)水硫酸鈉干燥后過(guò)濾。向?yàn)V液中加入乙酸乙酯(170 μL)和己烷(1.2 mL),將混合液冷卻至0℃,攪拌2小時(shí)以析出白色固體。通過(guò)過(guò)濾收集固體,依次用冰冷的己烷/乙酸乙酯(4:1,10 mL)、己烷/乙酸乙酯(9:1,10 mL)和己烷(10 mL,兩次)洗滌。最后,將所得白色固體在室溫下真空干燥,得到2,5-二氧代吡咯烷-1-基2-氯乙酸酯(563.9 mg,收率53%)。產(chǎn)物經(jīng)400 MHz NMR(CDCl3)表征,化學(xué)位移為δ 4.37(s,2H),2.87(s,4H)。
