以2-氨基-5-氟苯甲酸為原料合成2-氨基-3-溴-5-氟苯甲酸的一般步驟:將N-溴代琥珀酰亞胺(631 mg,3.55 mmol)分批加入到溶解于乙酸(5 mL)中的2-氨基-5-氟苯甲酸(7,500 mg,3.22 mmol)的攪拌溶液中,反應(yīng)混合物在室溫下攪拌過夜。通過過濾收集所得沉淀,用石油醚(20 mL)洗滌,并干燥至恒重,得到2-氨基-3-溴-5-氟苯甲酸(11,501 mg,產(chǎn)率66.4%),為透明米色固體。產(chǎn)物無需進(jìn)一步純化即可使用。產(chǎn)物表征數(shù)據(jù)如下:LCMS(t R = 1.35 min,純度= 100%),ESI m/z未檢測(cè)到離子;1H NMR(DMSO-d6)δ(ppm)6.76(寬單峰,2H),7.55(雙峰,J = 3.0 Hz,JH-F = 9.4 Hz,1H),7.71(雙峰,J = 3.0 Hz,JH-F = 7.8 Hz,1H),12.72(寬單峰,1H);13C NMR(DMSO-d6)δ(ppm)109.4(雙峰,JC-F = 7.1 Hz),111.1(雙峰,JC-F = 7.1 Hz),116.2(雙峰,JC-F = 22.1 Hz),124.9(雙峰,JC-F = 24.8 Hz),145.1,151.4(雙峰,JC-F = 235.3 Hz),168.1(雙峰,JC-F = 2.7 Hz)。
