以4-(芐氧基)-5-甲氧基-2-硝基苯甲醛為原料合成4-羥基-5-甲氧基-2-硝基苯甲醛的一般步驟:將4-(芐氧基)-5-甲氧基-2-硝基苯甲醛(10g,0.03mol)置于500mL圓底燒瓶中,加入200mL三氟乙酸溶解,若溶解不完全,可適量加入二氯甲烷助溶。反應(yīng)在室溫下進(jìn)行,持續(xù)攪拌過夜。通過薄層色譜(TLC,展開劑為二氯甲烷)監(jiān)測反應(yīng)進(jìn)度。反應(yīng)完成后,旋轉(zhuǎn)蒸發(fā)去除溶劑,殘余物用乙酸乙酯溶解。隨后,用1 mol/L NaOH溶液調(diào)節(jié)pH至1-10,以中和殘留的三氟乙酸,再用6 mol/L HCl溶液調(diào)節(jié)pH至約5,使苯酚鈉轉(zhuǎn)化為酚羥基。反應(yīng)液用乙酸乙酯萃取,合并有機(jī)相,用無水硫酸鈉干燥,然后濃縮至干。由于產(chǎn)物在正己烷中溶解性較差而雜質(zhì)溶解性較好,故用正己烷洗滌產(chǎn)物,干燥后得到5g黃色固體,即4-羥基-5-甲氧基-2-硝基苯甲醛,產(chǎn)率為72.9%。
