在氮?dú)獗Wo(hù)下,將7-甲氧基-3,4-二氫異喹啉-1(2H)-酮(19g,107mmol)溶解于二氯甲烷(250mL)中,并將溶液冷卻至-78℃。在1.5小時(shí)內(nèi),向該懸浮液中緩慢滴加三溴化硼(1M的二氯甲烷溶液,430mL,430mmol)。滴加完成后,保持反應(yīng)30分鐘,隨后移除冷卻浴,使反應(yīng)混合物逐漸升溫至室溫,并繼續(xù)攪拌過(guò)夜。反應(yīng)完成后,通過(guò)小心滴加甲醇(114mL,2.81mol)淬滅反應(yīng)混合物,將所得溶液攪拌1.5小時(shí)。隨后,將反應(yīng)混合物在減壓下濃縮。殘余物與甲醇共蒸發(fā)后,從水中結(jié)晶,經(jīng)過(guò)濾、水洗并干燥,得到目標(biāo)產(chǎn)物3,4-二氫-7-羥基-1(2H)-異喹啉酮(14.6g,收率81%)。產(chǎn)物的核磁共振氫譜(300MHz,DMSO-d6)數(shù)據(jù)如下:δH 9.47(s,1H),7.83(s,1H),7.25(d,J=2.7Hz,1H),7.09(d,J=8.2Hz,1H),6.85(dd,J=8.1,2.7Hz,1H),3.31(td,J=6.6,2.8Hz,2H),2.76(t,J=6.6Hz,2H)。
