將19.6 g (162.95 mmol)苯乙酮和5 g (54.32 mmol)草酰乙酸一水合物在100℃下攪拌反應(yīng)2小時(shí)。反應(yīng)完成后,將反應(yīng)液冷卻至40℃,隨后加入20 mL水和4 mL氨水。用50 mL二氯甲烷對(duì)混合物進(jìn)行兩次萃取。向得到的水相中加入2.64 mL (53.32 mmol)一水合肼,將混合物在100℃下繼續(xù)攪拌反應(yīng)2小時(shí)。反應(yīng)結(jié)束后,將反應(yīng)液冷卻至室溫。通過(guò)抽濾收集沉淀的晶體,用水洗滌后,在50℃的真空干燥箱中干燥過(guò)夜。最終得到4.3 g (24.97 mmol,產(chǎn)率15%) 6-苯基-3-噠嗪酮,為無(wú)色晶體。LC-MS (方法7): Rt = 1.39 min; m/z = 173 (M + H)+。1H-NMR (400 MHz, DMSO-d6, δ/ppm): 13.2 (s, 1H), 8.04 (d, 1H), 7.86 (d, 2H), 7.53-7.41 (m, 3H), 7.00 (d, 1H)。
