3-溴-2-甲氧基吡啶-4-胺(2)的合成:在0℃下,將2-甲氧基吡啶-4-胺(1, 20.0 g, 161.2 mmol)溶于二氯甲烷(200 mL)中。向此溶液中緩慢加入N-溴琥珀酰亞胺(26.6 g, 161.2 mmol),隨后將反應(yīng)混合物升溫至30℃并攪拌30分鐘。反應(yīng)完成后,用冰冷的水(100 mL)淬滅反應(yīng),并用二氯甲烷(300 mL)萃取反應(yīng)混合物。合并有機(jī)層,依次用水和鹽水洗滌,無(wú)水硫酸鈉干燥,過(guò)濾并減壓濃縮。粗產(chǎn)物用正戊烷和乙醚混合溶劑研磨,得到3-溴-2-甲氧基吡啶-4-胺(2),為黃色固體。產(chǎn)量:30.0 g,收率92%;MS(ESI)m/z 203.09 [M + 1]+。
