在室溫條件下,將2,4-二甲氧基苯甲醛(Sigma Aldrich; 1.1g,6.61mmol)溶解于20mL四氫呋喃(THF)中。向此溶液中逐滴加入2,4-二甲氧基芐胺(Sigma Aldrich; 1.5mL,9.98mmol),隨后加入三乙酰氧基硼氫化鈉(Sigma Aldrich; 17g; 80.4mmol)。反應(yīng)混合物逐漸變得混濁,于室溫下攪拌反應(yīng)過夜。反應(yīng)完成后,用飽和碳酸氫鈉(NaHCO3)溶液稀釋反應(yīng)混合物,隨后用乙酸乙酯(EtOAc)進(jìn)行萃取。合并有機(jī)相,用無(wú)水硫酸鎂(MgSO4)干燥,過濾后減壓濃縮。所得粗產(chǎn)物通過硅膠柱色譜法純化(使用40g ISCO柱,洗脫梯度為0至100%乙酸乙酯/己烷),得到雙(2,4-二甲氧基芐基)胺(2.08g,6.55mmol,產(chǎn)率99%,純度96%)。
