以乙醇和2-氯吡啶-5-乙酸為原料合成2-(6-氯吡啶-3-基)乙酸乙酯的一般步驟如下:向6-氯-3-吡啶乙酸(1g,5.83mmol)的乙醇溶液中緩慢加入濃硫酸(1.6mL)。將反應(yīng)混合物加熱至回流狀態(tài),保持4小時(shí)。反應(yīng)完成后,將混合物冷卻至室溫,并在旋轉(zhuǎn)蒸發(fā)儀中濃縮以去除大部分乙醇。將濃縮后的殘余物用乙酸乙酯溶解,隨后用飽和碳酸氫鈉溶液洗滌以中和殘余的酸。有機(jī)相經(jīng)無(wú)水硫酸鎂干燥后,過(guò)濾并減壓濃縮,得到粗產(chǎn)物。最后,通過(guò)硅膠柱色譜法純化粗產(chǎn)物,得到2-(6-氯吡啶-3-基)乙酸乙酯(1.1g,收率95%)。
