以4,5,6,7-四氫苯并[B]噻吩-3-羧酸乙酯為原料合成4,5,6,7-四氫苯并[b]噻吩-3-羧酸的一般步驟如下:通用方法:將氫氧化鉀(KOH,1.7 g,30 mmol)加入到4,5,6,7-四氫苯并[B]噻吩-3-羧酸乙酯(30 mmol)的水(H2O,20 mL)乳液中。在劇烈攪拌下加熱混合物直至獲得澄清溶液(約1小時),然后繼續(xù)攪拌30分鐘。將反應溶液冷卻至室溫,用甲苯(PhMe)洗滌;分離水層并用鹽酸(HCl)酸化至pH值為4。過濾收集沉淀,并用乙醇(EtOH)-N,N-二甲基甲酰胺(DMF)混合溶劑進行重結晶。
