以2-甲氧基-4-氟苯腈為原料合成4-氟-2-羥基苯腈的一般步驟:將4-氟-2-甲氧基芐腈(4.53 g,30 mmol)與三氯化鋁(5.0 g,37.6 mmol;購自Aldrich)在無水甲苯(30 mL)中的混合物于130℃下攪拌反應(yīng)18小時(shí)。反應(yīng)完成后,將混合物冷卻至室溫,加入冰水(約50 mL),用乙醚(20 mL × 2)萃取。合并有機(jī)相,依次用水和飽和鹽水洗滌,然后用無水硫酸鎂干燥。減壓濃縮后,得到4-氟-2-羥基苯腈(3.90 g,28.5 mmol,收率95%),為白色固體。其結(jié)構(gòu)通過以下表征確認(rèn):1H NMR(DMSO-d6, 300 MHz)δ ppm:6.74-6.84(2H,m,Ar-H),7.71(1H,dd,J = 7 Hz,J = 8.5 Hz,Ar-H),11.64(1H,s,OH);13C NMR(DMSO-d6, 75.5 MHz)δ ppm:95.13, 102.45, 102.78, 106.53, 106.83, 115.53, 134.68, 134.84, 161.41, 161.58, 163.00, 166.35。高分辨質(zhì)譜(ESI-)計(jì)算值C7H3NOF(M-H)為136.0199,實(shí)測值為136.0199。
