以茴香烯為原料合成5-(4-羥基苯基)-3H-1,2-二硫-3-硫酮的一般步驟如下:將茴香烯(1)(32.5 g, 0.21 mol)與硫粉(45 g, 1.40 mol)在N,N-二甲基甲酰胺(250 mL)中混合,加熱回流反應(yīng)8小時(shí)。反應(yīng)完成后,減壓蒸餾除去溶劑,所得殘余物用甲苯溶解。隨后,用2N氫氧化鈉水溶液對甲苯溶液進(jìn)行萃取,得到橙色固體沉淀(8.5 g)。將該固體產(chǎn)物溶于沸水中,緩慢加入鹽酸至酸性,析出橙色沉淀(2),收率50%,熔點(diǎn)205℃(文獻(xiàn)值188-189℃)。產(chǎn)物的結(jié)構(gòu)通過1H NMR(DMSO-d6)和質(zhì)譜(ESI)確認(rèn):1H NMR (DMSO-d6) δ 6.86 (d, 2H), 7.68 (s, 1H), 7.75 (d, 2H), 10.51 (s, 1H, -OH); MS (ESI) m/z 225 [M-H]-。
