以4-溴-2-硝基苯胺為原料合成2-碘-4-溴-6-硝基苯胺的一般步驟:將4-溴-2-硝基苯胺(14.3 g,0.0659 mol)一次性加入溶解于乙醇(300 mL)中的碘(17.6 g,0.0692 mol)溶液中,隨后加入硫酸銀(I)(20.4 g,0.0659 mol)。反應(yīng)混合物在室溫下攪拌18小時(shí)后,進(jìn)行過濾。所得固體用二氯甲烷洗滌,直至所有橙色產(chǎn)物溶解。合并濾液,經(jīng)真空蒸發(fā)后得到的固體用乙醚/40-60石油醚(1:1)混合溶劑洗滌,過濾后得到4-溴-2-碘-6-硝基苯胺,為橙色固體(19.8 g,收率88%),無需進(jìn)一步純化即可用于后續(xù)反應(yīng)。1H NMR(400 MHz,CDCl3,δ):6.15(2H,br s),8.00(1H,s),8.42(1H,s)。
