通用方法:在攪拌條件下,向3-氰基吲哚(15 mmol)的無水乙醇溶液中加入40% NaOH水溶液(5 mL)。反應過程通過薄層色譜(TLC)監(jiān)測。隨后,緩慢向反應混合物中滴加30% H2O2水溶液(10.5 mL, 104.7 mmol)。反應完成后,過濾收集沉淀的固體,用水洗滌,并用無水乙醇進行重結晶。最后,將產(chǎn)物在真空下干燥,得到1H-吲哚-3-羧酰胺(20a)。產(chǎn)物為白色固體,收率75%,熔點195-197℃(文獻值197-199℃[31])。1H NMR (δ, ppm): 6.84 (s, 1H, NH), 7.13-7.21 (m, 2H, ArH), 7.47 (d, J = 8.0 Hz, 2H, ArH), 8.08 (s, 1H, NH), 8.20 (d, J = 7.5 Hz, 1H, ArH), 11.60 (s, 1H, NH)。
