2-氟-3-甲氧基吡啶(3)的合成:在100 mL丙酮中,將2-氟吡啶-3-醇(10 mmol)、碘甲烷(20 mmol)和碳酸鉀(20 mmol)混合,回流反應(yīng)過夜。反應(yīng)完成后,過濾除去無機(jī)鹽,并通過旋轉(zhuǎn)蒸發(fā)去除溶劑。粗產(chǎn)物通過柱層析純化(洗脫劑比例為乙酸乙酯:己烷=1:5),得到無色液體產(chǎn)物[19],收率為95%。產(chǎn)物表征數(shù)據(jù)如下:1H NMR(CDCl3)δ 7.74-7.75(m, 1H, C6-H),7.26-7.31(m, 1H, C4-H),7.11-7.15(m, 1H, C5-H),3.91(s, 3H, OMe);19F NMR(CDCl3)δ 90.15(s);ESI-MS:128(M + 1)+。
