制備例1 1-(2-氯-4-硝基苯基)-4-甲基哌嗪的合成:在干燥的反應(yīng)燒瓶中,將3-氯-4-氟硝基苯(10.05g,57.2mmol)與N-甲基哌嗪(17.2g,172mmol)混合。反應(yīng)過程中觀察到放熱現(xiàn)象,持續(xù)攪拌1小時后,向反應(yīng)混合物中加入水進行淬滅。通過過濾收集生成的棕色固體沉淀,并用大量水洗滌以去除未反應(yīng)的原料和副產(chǎn)物。最后,將產(chǎn)物在空氣中干燥,得到1-(2-氯-4-硝基苯基)-4-甲基哌嗪(14.1g,收率96%)。產(chǎn)物經(jīng)1H NMR(400MHz,DMSO-d6)和質(zhì)譜(MS)表征確認(rèn):1H NMR δ 2.24(s,3H),2.43-2.55(m,4H),3.11-3.24(m,4H),7.28(d,J = 9.03Hz,1H),8.14(dd,J = 9.03,2.76Hz,1H),8.21(d,J = 2.51Hz,1H);MS m/z 255(M+)。
