步驟3:在氮?dú)獗Wo(hù)下,向100 mL三頸圓底燒瓶中加入5-烯丙基嘧啶-4,6-二醇(4 g)和磷酰氯(25 mL)。將反應(yīng)混合物加熱至80℃,并在此溫度下攪拌反應(yīng)3小時(shí)。反應(yīng)完成后,將混合物冷卻至室溫,隨后在減壓條件下除去過量磷酰氯。將殘余物用飽和碳酸氫鈉溶液中和,調(diào)節(jié)pH至8-9。用二氯甲烷(100 mL)稀釋水層,并用二氯甲烷(2×50 mL)進(jìn)行萃取。合并有機(jī)層,用飽和食鹽水(50 mL)洗滌,并用無水硫酸鈉干燥。減壓濃縮有機(jī)相,得到粗產(chǎn)物。通過柱色譜法純化粗產(chǎn)物,使用甲醇/氯仿(0.4:9.6,v/v)作為洗脫劑,最終獲得5-烯丙基-4,6-二氯嘧啶(2.7 g)。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(DMSO-d6)確認(rèn):δ 3.59-3.61(d,2H),5.00-5.05(dd,1H),5.13-5.16(dd,1H),5.85-5.94(m,1H),8.8(s,1H)。
