以N-(6-溴-1-氧代-2,3-二氫-1H-茚-5-基)乙酰胺為原料合成5-氨基-6-溴-2,3-二氫-1H-茚-1-酮的一般步驟:將N-(6-溴-1-氧代-2,3-二氫-1H-茚-5-基)乙酰胺(13.1 g,48.9 mmol)與6 M鹽酸水溶液(260 mL,1.560 mol)混合,在100℃下攪拌反應(yīng)1小時(shí)。反應(yīng)完成后,將反應(yīng)液冷卻至0℃,用10 M氫氧化鈉水溶液調(diào)節(jié)pH至8。析出的固體經(jīng)過濾收集,用水洗滌,真空干燥,得到淺棕色粉末狀產(chǎn)物5-氨基-6-溴-2,3-二氫-1H-茚-1-酮(10.8 g,收率98%)。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(400 MHz,CDCl3)確認(rèn):δ 7.86(s,1H),6.74(s,1H),4.68(s,2H),2.98(t,J = 6.0 Hz,2H),2.64(t,J = 6.0 Hz,2H)。
