一般步驟:向圓底燒瓶中加入4-氟-2-異丙氧基-1-硝基苯(49.3 g,248 mmol)、乙醇(600 mL)和10% Pd/C催化劑(50%水溶液,Degussa型E101;10 g,相當(dāng)于起始硝基化合物質(zhì)量的20%)。將燒瓶用橡膠隔膜密封,進(jìn)行三次抽真空-氫氣回填循環(huán)以脫氣。在氫氣氛圍下(使用氫氣氣球維持壓力),室溫?cái)嚢璺磻?yīng)混合物4天。反應(yīng)完成后,將混合物通過(guò)硅藻土墊過(guò)濾,并用甲醇充分洗滌硅藻土墊。合并濾液與洗滌液,減壓濃縮至干,得到棕色液體4-氟-2-異丙氧基苯胺(41.0 g,收率98%),該產(chǎn)物無(wú)需進(jìn)一步純化即可用于后續(xù)反應(yīng)。產(chǎn)物表征數(shù)據(jù)如下:1H NMR (300 MHz, DMSO-d6) δ 6.67-6.72 (dd, 1H, J = 2.7, 11.0 Hz), 6.55-6.60 (dd, 1H, J = 6.0, 8.7 Hz), 6.43-6.49 (td, 1H, J = 2.7, 8.4 Hz), 4.47-4.55 (m, 3H), 1.23-1.25 (d, 6H, J = 6.0 Hz); 19F NMR (282 MHz, DMSO-d6) δ -127.4 (sextet); MS (ESI) m/z = 170 [M+H]+.
