以2-碘芴和碘甲烷為原料合成2-碘-9,9-二甲基芴的一般步驟:將2-碘芴(3g,10.27mmol,1當(dāng)量)溶解于50mL無水四氫呋喃中。將溶液冷卻至0℃后,加入2.6g叔丁醇鉀(23.16mmol,3當(dāng)量),此時(shí)透明溶液轉(zhuǎn)變?yōu)榧t色。攪拌10分鐘后,加入1.44g碘甲烷(23.16mmol,3當(dāng)量),紅色溶液變?yōu)槿闋?。將反?yīng)混合物從0℃緩慢升溫至室溫,并繼續(xù)攪拌12小時(shí)。反應(yīng)完成后,加入100mL去離子水,用乙酸乙酯(100mL×3)進(jìn)行萃取。合并有機(jī)相,用硫酸鎂干燥后過濾。濾液在減壓下濃縮,得到粘稠液體。通過硅膠柱色譜法(洗脫劑:純正己烷)進(jìn)一步純化,得到3.0g目標(biāo)產(chǎn)物2-碘-9,9-二甲基芴,產(chǎn)率為91%。1H NMR(300MHz,CDCl3,δ):1.47(s,6H),7.34-7.36(m,2H),7.40-7.45(m,1H),7.48(s,1H),7.64-7.70(m,2H),7.75-7.76(m,1H)。
