熒光顯影染料
ToTo-1是高靈敏核酸顯影劑,常用于細(xì)胞核顯影。ToTo-1是TO-PRO-1的二連體(dimer),屬雙菁類(Cyanine dimers)熒光探針。雙菁類探針由兩個單菁染料分子(一般來自托普家族,TO-PRO family)對稱鏈接而成,共帶4個正電荷。這個結(jié)構(gòu)大大增強(qiáng)了其對DNA的親和性,親和能力提高可達(dá)100倍。另外,雙菁類探針本身沒有熒光,但是嵌入DNA后熒光增強(qiáng)100-1000倍,屬已知最強(qiáng)最亮的一類核酸顯影劑之一,它們的靈敏度甚至達(dá)到可以顯影單個DNA鏈(Science 264, 822 (1994))。雙菁類探針也包含10個家族成員,它們的顯影熒光也可覆蓋所有可見光光譜范圍,從456nm(PoPo-1)到660nm(ToTo-3),因此從它們中總能找到一個譜段合適的探針。雙菁類探針難于透過細(xì)胞膜,適用于已固定細(xì)胞的細(xì)胞核對染(counterstaining),可用于流式細(xì)胞儀或者熒光顯微鏡實驗。雙菁類探針也用于瓊脂糖膠核酸顯影。
ToTo-1與核酸或細(xì)胞核結(jié)合后顯綠色熒光(ex/em ~ 514/533 nm)。
