a)1-甲基-1H-吲哚-4-甲醛的合成:在氬氣保護(hù)下,向1H-吲哚-4-甲醛(413 mg,2.85 mmol)的無水N,N-二甲基甲酰胺(DMF,6.5 mL)溶液中分批加入氫化鈉(171 mg,60%油分散體,4.27 mmol)。反應(yīng)混合物于室溫下攪拌40分鐘。隨后,緩慢滴加碘甲烷(0.36 mL,5.78 mmol),繼續(xù)在室溫下攪拌反應(yīng)12小時(shí)。反應(yīng)完成后,加入水(25 mL)淬滅反應(yīng),并用乙酸乙酯(3×25 mL)萃取。合并有機(jī)相,依次用水(20 mL)和飽和食鹽水(20 mL)洗滌,無水硫酸鈉干燥,過濾并減壓濃縮,得到黃色油狀粗產(chǎn)物。通過硅膠柱色譜(洗脫劑:二氯甲烷)純化,得到1-甲基-1H-吲哚-4-甲醛(452 mg,收率99%)為黃色油狀物。1H NMR(400 MHz,DMSO-d6)δ 10.20(s, 1H),7.84(d, J = 8.0 Hz, 1H),7.68(d, J = 7.2 Hz, 1H),7.58(d, J = 2.8 Hz, 1H),7.38-7.34(m, 1H),7.08(d, J = 3.2 Hz, 1H),3.87(s, 3H)。
