向裝有攪拌子的20 mL螺旋蓋小瓶中加入N,N-雙((1-(叔丁基)-1H-1,2,3-三唑-4-基)甲基)丙-2-炔-1-胺(109 mg,0.33 mmol,1.0當(dāng)量)和疊氮乙酸(50 mg,0.50 mmol,1.5當(dāng)量)于3 mL四氫呋喃(THF)中。隨后向混合物中加入N,N'-二異丙基乙胺(68 mg,0.53 mmol,1.6當(dāng)量)和三(三苯基膦)溴化銅(I)(31 mg,0.033 mmol,10 mol%)。將反應(yīng)混合物密封并在60°C下劇烈攪拌過夜。反應(yīng)完成后,向混合物中加入2 mL水和約0.2 g CupriSorb?,繼續(xù)攪拌30分鐘后過濾。將濾液減壓濃縮,所得粗產(chǎn)物通過快速柱色譜法純化(25 g硅膠,洗脫劑為含1%乙酸的40%甲醇/乙酸乙酯溶液,Rf = 0.2,KMnO4顯色),得到109 mg目標(biāo)產(chǎn)物(產(chǎn)率76%),為白色固體。1H NMR(300 MHz,D2O)δ 8.22(s,2H),8.14(s,1H),5.10(s,2H),4.72(s,2H),4.39(s,4H),1.66(s,18H)。
