以2-羥基吡啶-4-羧酸甲酯為原料合成4-(羥甲基)吡啶-2(1H)-酮的一般步驟:向2-甲氧基-1,2-二氫-4-吡啶羧酸甲酯(1.37 g,8.98 mmol)的無水四氫呋喃(23 mL)懸浮液中緩慢滴加2 M硼氫化鋰/四氫呋喃溶液(22.5 mL,45 mmol),在氮?dú)獗Wo(hù)下將反應(yīng)混合物加熱至55℃并維持3.5小時。反應(yīng)完成后,小心加入甲醇(15 mL)和水(3 mL),隨后在室溫下攪拌30分鐘。將混合物濃縮后,再次小心加入甲醇(10 mL)。繼續(xù)攪拌30分鐘后,將混合物吸附于硅膠上,并通過硅膠柱層析純化,使用0至30%甲醇-二氯甲烷梯度洗脫,得到目標(biāo)產(chǎn)物4-(羥甲基)吡啶-2(1H)-酮,為灰白色固體(0.99 g,收率88%)。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(400 MHz,DMSO-d6)確認(rèn):δ 11.29(br s,1H),7.23(d,J = 6.7 Hz,1H),6.21(s,1H),6.03(dd,J = 6.7, 1.3 Hz,1H),5.27(t,J = 5.9 Hz,1H),4.28(d,J = 5.9 Hz,2H)。
