以1-Cbz-2-Boc-1-甲基肼為原料合成1-Boc-2-甲基肼的一般步驟如下:在室溫和標(biāo)準(zhǔn)大氣壓下,將24.8g(70.8mmol)1-芐基-2-叔丁基-1-甲基肼-1,2-二甲酸酯溶解于500mL乙醇中,加入1.24g鈀(10%活性炭)作為催化劑進(jìn)行氫化反應(yīng)。反應(yīng)完成后,將反應(yīng)混合物通過硅藻土過濾以去除催化劑,濾液在減壓條件下濃縮并干燥,得到產(chǎn)物。產(chǎn)量為12.3克,純度為48%,占理論產(chǎn)率的57%。
