以硝基乙酸乙酯和2-丙炔-1-醇為原料合成5-羥甲基異噁唑-3-甲酸乙酯的一般步驟如下:中間體4-制備5-(羥甲基)異惡唑-3-羧酸乙酯。在Ace壓力管中,將2-丙炔-1-醇(1.0 mL,16.76 mmol)和硝基乙酸乙酯(3.79 mL,33.52 mmol)溶于乙醇(23.5 mL)中,隨后加入1,4-二氮雜雙環(huán)[2.2.2]辛烷(DABCO,0.194 g,1.68 mmol)。密封反應(yīng)管,于80℃加熱反應(yīng)72小時。反應(yīng)完成后,冷卻至室溫,將反應(yīng)混合物減壓濃縮至干。所得殘余物通過硅膠快速柱色譜法純化,洗脫劑為二氯甲烷與甲醇的混合溶液(甲醇體積分?jǐn)?shù)0%至6%),最終得到5-(羥甲基)異惡唑-3-羧酸乙酯2.32 g,收率81%,為油狀物。ESI/APCI(+):m/z 172([M+H]+)。
