以3-芐基-6-叔丁基-3,6-二氮雜雙環(huán)[3.2.0]庚烷-3,6-二羧酸酯(2.61 mmol)為原料,在Parr瓶中將其溶于甲醇(0.1 M)。加入鈀/碳催化劑(Johnson Mathey,5% wt,61.1% H2O,60 mg)。將混合物置于Parr振蕩器上,通過真空除去空氣。用氮?dú)夥磸?fù)吹掃系統(tǒng),隨后在15 PSI下振蕩約1.5小時(shí)。通過TLC監(jiān)測反應(yīng)進(jìn)度,確認(rèn)原料完全消耗。反應(yīng)完成后,將混合物通過硅藻土墊過濾。濃縮濾液,得到粘稠的透明殘余物,即3,6-二氮雜雙環(huán)[3.2.0]庚烷-6-羧酸叔丁酯,收率為66.1%。該產(chǎn)物純度足夠高,無需進(jìn)一步純化即可用于后續(xù)反應(yīng)。質(zhì)譜(ESI)計(jì)算值:C10H18N2O2,198.26;實(shí)測值:m/z 199.2 [M + H]+。1H NMR(400 MHz,CDCl3)δ 4.73-4.50(m,1H),3.98(t,J = 8.3 Hz,1H),3.46-3.15(m,2H),3.09-2.99(m,1H),2.93-2.82(m,1H),2.73-2.62(m,1H),2.53-2.42(m,1H),2.31(s,1H),1.43(s,9H)。
