步驟1:4-溴-2-(二氟甲基)吡啶的合成:在室溫下,向4-溴吡啶-2-甲醛(5.0 g,26.88 mmol)中加入二乙氨基三氟化硫(7.03 mL,53.7 mmol),并將反應(yīng)混合物攪拌16小時。反應(yīng)完成后,用碳酸氫鈉水溶液(50 mL)淬滅反應(yīng),隨后用乙酸乙酯(3×100 mL)萃取。合并有機(jī)層,用鹽水(2×50 mL)洗滌,經(jīng)無水硫酸鈉干燥,過濾并濃縮。通過硅膠柱色譜(100-200目)純化,使用5%乙酸乙酯的己烷溶液作為洗脫劑,得到4-溴-2-(二氟甲基)吡啶(3.0 g,14.492 mmol,54%收率)。1H NMR(400 MHz,CDCl3)δ= 8.48(d,J = 4.9 Hz,1H),7.82(d,J = 1.5 Hz,1H),7.66-7.50(m,1H),6.60(t,1H)。LCMS:207.9 [M + H]+。
