以4-氯-1H-吡唑并[4,3-c]吡啶(1.5 g,9.77 mmol,1.00當(dāng)量)為起始原料,將其溶解于1,4-二惡烷(25 mL)中。隨后,加入氫氧化鉀(2.0 g,35.65 mmol,3.60當(dāng)量),并在75℃下攪拌反應(yīng)混合物。在此溫度下,緩慢加入碘(4.95 g,19.50 mmol,2.00當(dāng)量),反應(yīng)持續(xù)4小時。反應(yīng)完成后,用飽和硫代硫酸鈉五水合物水溶液淬滅反應(yīng),通過過濾收集生成的固體。最終得到4-氯-3-碘-1H-吡唑并[4,3-c]吡啶(2.5 g,收率92%),為淺黃色固體。產(chǎn)物經(jīng)LC-MS(ES,m/z)分析,顯示分子離子峰為280 [M + H]。
