向1H-咪唑-2-甲醛(480 mg,5 mmol)和無水碳酸鉀(936 mg,6 mmol)在N,N-二甲基甲酰胺(7 mL)中的懸浮液中加入碘乙烷(829 mg,6 mmol)。將反應(yīng)混合物在50℃下攪拌加熱5小時。反應(yīng)完成后,減壓蒸餾除去溶劑。將殘余物用蒸餾水(30 mL)和乙酸乙酯(30 mL)進行液液分配。水相用乙酸乙酯(20 mL×3)進一步萃取。合并所有有機相,用飽和氯化鈉溶液洗滌,無水硫酸鈉干燥,減壓濃縮,得到1-乙基-1H-咪唑-2-甲醛,為淺黃色油狀物(520 mg,收率84%)。產(chǎn)物經(jīng)1H NMR(400 MHz,CDCl3)表征:δ 1.42-1.46(t,J = 7.2 Hz,3H),4.42-4.47(q,2H),7.19(s,1H),7.29(s,1H),9.82(s,1H)。LC-MS(ESI)m/z:125(M + 1)+。
