以2-氨基-5-甲基噻吩-3-甲酸乙酯(2.3 g,12.0 mmol)和甲酰胺(28 mL)為原料,合成6-甲基-3H-噻吩[2,3-d]嘧啶-4-酮的一般步驟:將2-氨基-5-甲基噻吩-3-甲酸乙酯和甲酰胺的懸浮液在180℃下加熱反應(yīng)16小時(shí)。反應(yīng)完成后,將反應(yīng)混合物用100 mL水淬滅,并繼續(xù)攪拌30分鐘。通過過濾收集沉淀的固體,用戊烷洗滌,經(jīng)抽濾干燥后,得到6-甲基-3H-噻吩[2,3-d]嘧啶-4-酮(1.0 g,粗品),為灰白色固體。質(zhì)譜分析結(jié)果:計(jì)算值(M + H):167.02;實(shí)測(cè)值(M + H):167.1。
