實(shí)施例4 2-(甲硫基)嘧啶-4-甲醛(4)的合成:在60℃條件下,將4-(二甲氧基甲基)-2-(甲硫基)嘧啶(3)(53.7g, 268mmol)緩慢加入至1.2N HCl水溶液(300mL, 268mmol, 1.0當(dāng)量)中,持續(xù)攪拌3小時(shí)。反應(yīng)完成后,將混合物冷卻至室溫,并通過緩慢加入固體碳酸氫鈉進(jìn)行中和。隨后,用乙醚(3×150mL)對(duì)粗產(chǎn)物進(jìn)行萃取,合并有機(jī)層后在減壓條件下濃縮,得到目標(biāo)產(chǎn)物2-(甲硫基)嘧啶-4-甲醛(4),為黃色固體(14.2g, 產(chǎn)率34%)。LC-MS m/z 155(M+1)。參考文獻(xiàn):WO 2006/009734 A1, 第67頁(yè)。
