以4-乙烯基苯甲酸為原料合成(4-乙烯基苯基)甲醇的一般步驟如下:按照文獻[31]所述方法,將4-乙烯基苯甲酸(500 mg,3.37 mmol)溶于Et2O(25 mL)中,緩慢滴加至0℃的LiAlH4(758 mg,19.97 mmol)在THF(80 mL)中的懸浮液。反應(yīng)混合物逐漸升溫至室溫并持續(xù)攪拌1小時。反應(yīng)完成后,依次緩慢加入水(256 μL)、10 wt% NaOH溶液(512 μL)和水(768 μL)以淬滅反應(yīng)?;旌衔锝?jīng)劇烈攪拌直至白色固體形成。粗反應(yīng)混合物經(jīng)過濾,上清液用無水Na2SO4干燥后,減壓濃縮。所得油狀物通過硅膠柱色譜法純化,洗脫劑為己烷:EtOAc(4:1),得到目標產(chǎn)物(4-乙烯基苯基)甲醇(280 mg,62%),為無色油狀物。產(chǎn)物的物理和光譜數(shù)據(jù)與文獻[31]報道一致。1H NMR(CDCl3, 500 MHz)δ:4.66(s, 2H), 5.24(d, J = 11.5 Hz, 1H), 5.73(d, J = 18.5 Hz, 1H), 6.69-6.75(dd, J = 11.0, 6.5 Hz, 1H), 7.31(d, J = 8.0 Hz, 2H), 7.39(d, J = 8.0 Hz, 2H)。
