以2-氯-5-氟煙腈為原料合成5-氟-1H-吡唑并[3,4-b]吡啶-3-胺的一般步驟如下:向2-氯-5-氟煙腈(50g,321.7mmol)的1-丁醇(1L)溶液中加入一水合肼(150mL,3.2mol),并將混合物回流4小時。反應(yīng)完成后,將混合物冷卻至室溫并濃縮。所得沉淀依次用水(2次)和乙醚(2次)洗滌,隨后在真空下干燥過夜,得到5-氟-1H-吡唑并[3,4-b]吡啶-3-胺(44g,產(chǎn)率88%),為黃色固體。1H NMR(DMSO-d6, 300MHz)數(shù)據(jù)如下:δ5.53(s,2H);7.94(dd,1H);8.35(dd,1H);12.02(s,1H)。
