5-甲基-3-吡啶甲醇的制備:按照2:2:1的摩爾比將氯化鎂、硼氫化鉀與四氫呋喃混合,升溫至67℃,回流2小時(shí),隨后冷卻至室溫,得到溶液B。將S1步驟中制得的5-甲基煙酸甲酯溶于四氫呋喃中,調(diào)節(jié)溫度至40℃,緩慢滴加溶液B,控制滴加時(shí)間在1.3小時(shí)內(nèi)完成。滴加完畢后,保持40℃繼續(xù)反應(yīng)1小時(shí),期間持續(xù)攪拌。反應(yīng)完成后,冷卻至室溫,得到溶液C。向溶液C中加入甲醇以淬滅反應(yīng),隨后升溫至40℃,通過(guò)旋轉(zhuǎn)蒸發(fā)去除四氫呋喃和甲醇,得到濃縮物D。向濃縮物D中加入水,用乙酸乙酯進(jìn)行萃取,共萃取三次。合并乙酸乙酯相,加入無(wú)水硫酸鈉干燥至水分含量為0.3wt%,過(guò)濾后,取乙酸乙酯相進(jìn)行旋轉(zhuǎn)蒸發(fā),最終得到5-甲基-3-吡啶甲醇。
