以5-硝基酞嗪為起始原料合成5-氨基酞嗪的一般步驟:在50℃條件下,將0.88g(5.0mmol)5-硝基酞嗪溶解于30mL四氫呋喃中,隨后加入4.37g(25.1mmol)亞硫酸氫鈉溶于15mL水的溶液。將反應(yīng)混合物在50℃下持續(xù)攪拌15分鐘。反應(yīng)完成后,用乙酸乙酯(3×100mL)萃取有機相,合并有機相后用鹽水(50mL)洗滌,并用無水硫酸鎂干燥。過濾后,減壓濃縮除去溶劑,得到亮黃色沉淀的粗產(chǎn)物。將粗產(chǎn)物在熱甲醇中重結(jié)晶,最終得到0.49g(產(chǎn)率67%)純化的5-氨基酞嗪。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(400MHz,DMSO-d6)和13C NMR(100.17MHz,DMSO-d6)確認(rèn),具體數(shù)據(jù)如下:1H NMR δ:9.80(s,1H),9.60(s,1H),7.60(dd,J = 4.6,7.6Hz,1H),7.20(dd,J = 1.9,4.6Hz,1H),7.02(d,J = 7.6Hz,1H),6.50(s,2H);13C NMR δ:150.1,146.5,145.1,133.2,126.1,114.7,114.1,112.2。
