步驟b)2-氯-4-(氯甲基)吡啶的合成:將11.3 g步驟a)中獲得的(2-氯吡啶-4-基)甲醇溶解于200 mL二氯甲烷中,向該溶液中緩慢加入6.896 mL亞硫酰氯,隨后加入2.1 mL二甲基甲酰胺作為催化劑。將反應(yīng)混合物在室溫下攪拌3小時(shí)。反應(yīng)完成后,緩慢滴加50 mL水以淬滅反應(yīng)。分離有機(jī)層,用無(wú)水硫酸鎂干燥,過(guò)濾除去干燥劑,濾液經(jīng)減壓濃縮,得到12.8 g(產(chǎn)率100%)2-氯-4-(氯甲基)吡啶,產(chǎn)物為琥珀色液體,無(wú)需進(jìn)一步純化即可用于后續(xù)反應(yīng)。薄層色譜(TLC)分析顯示Rf值為0.44(展開(kāi)劑:二氯甲烷)。
