在制備例1中獲得的(6-甲氧基吡啶-3-基)甲醇(2)(537.8g,3.86mol)溶解于二甲基甲酰胺(1.6L)中,于氮氣保護下,冰浴冷卻,1.3小時內(nèi)緩慢滴加亞硫酰氯(310mL,4.25mol)。滴加完畢后,繼續(xù)在冰浴中攪拌反應(yīng)混合物1小時。隨后,在溫度不超過23℃的條件下,依次向反應(yīng)混合物中加入甲苯(5.4L)和2N氫氧化鈉水溶液(5.4L)。攪拌約10分鐘后,分離水層,有機層用水(2.7L)洗滌。減壓濃縮有機層至干,殘余物中加入甲苯(1.0L)進行共沸蒸餾,最終得到618.8g目標產(chǎn)物5-氯甲基-2-甲氧基吡啶(3),為淺黃色油狀物(含量556.3g,收率91.4%)。產(chǎn)物結(jié)構(gòu)經(jīng)1H-NMR(CDCl3)確認:δ(ppm)8.15(1H,d,J=2.4Hz),7.63(1H,dd,J=2.4Hz,8.4Hz),6.75(1H,d,J=8.4Hz),4.55(2H,s),3.94(3H,s)。
