制備7:4-碘甲基四氫吡喃; 將制備6(4-甲磺酰氧甲基四氫吡喃,328g,1.69mol)和碘化鈉(507g,3.4mol)在丙酮(3.3L)中的混合物加熱回流4小時。通過薄層色譜(TLC,展開劑:乙醚)監(jiān)測反應(yīng)進(jìn)程,發(fā)現(xiàn)仍有大量甲磺酸酯未反應(yīng),遂補(bǔ)加碘化鈉(127g,0.65mol)并繼續(xù)回流16小時。反應(yīng)完成后,將反應(yīng)混合物冷卻至室溫并過濾。濾餅用丙酮洗滌,干燥后,將所得固體溶于乙醚(800mL)和水(800mL)的混合溶劑中進(jìn)行分配萃取。水相再用乙醚(200mL)萃取一次,合并所有乙醚萃取液,用10%硫代硫酸鈉溶液(300mL)洗滌以去除碘的顏色,隨后用水(300mL)洗滌。有機(jī)相經(jīng)無水硫酸鎂干燥后,減壓濃縮除去溶劑,得到目標(biāo)產(chǎn)物4-碘甲基四氫吡喃(365g,收率92%)。產(chǎn)物的核磁共振(NMR)譜圖與預(yù)期結(jié)構(gòu)相符。
