以2-氟-5-甲氧基-4-硝基苯甲酸為原料合成4-氨基-2-氟-5-甲氧基苯甲酸的一般步驟:將2-氟-5-甲氧基-4-硝基苯甲酸(10g,46mmol)溶于HOAc(50mL)和MeOH(50mL)的混合溶液中,在氫氣球氣氛下進行氫化反應(yīng),反應(yīng)過夜。反應(yīng)完成后,將反應(yīng)液通過硅藻土過濾,濾液濃縮至干,得到殘余物。殘余物用乙醚和乙酸乙酯混合溶劑研磨,析出固體。過濾收集固體,干燥后得到4-氨基-2-氟-5-甲氧基苯甲酸,為淺黃色粉末(8.3g,收率98%)。產(chǎn)物經(jīng)1H NMR(400MHz,DMSO-d6)表征:δ 3.76(s,3H),5.88(br s,2H),6.36(d,J = 16Hz,1H),7.13(d,J = 8Hz,1H),12.32(br s,1H)。質(zhì)譜分析:[M + H]+計算值(C8H8FNO3)為186,實測值為186。
