Step 2: Preparation of 6-bromo-3-methoxypyridin-2-amine
A mixture of 6-bromo-3-methoxy-2-nitropyridine (14.4 g, 61.8 mmol), acetic acid (200 mL), and iron powder (14.0 g, 0.25 mol) was stirred and reacted at 90°C for 1.5 hours. Upon completion of the reaction, the mixture was cooled to room temperature and diluted with ethyl acetate. Subsequently, the mixture was filtered through diatomaceous earth and the filtrate was concentrated under vacuum. Purification by fast column chromatography using dichloromethane as eluent afforded the target compound 6-bromo-3-methoxypyridin-2-amine (10.2 g, 50.2 mmol) as a white solid.1H NMR (400 MHz, DMSO-d6) δ: 3.81 (s, 3H), 4.78 (br.s, 2H), 6.73 (d, J = 7.8 Hz 1H). | [References]
